A communal catalogue reveals Earth’s multiscale microbial diversity

Our growing awareness of the microbial world's importance and diversity contrasts starkly with our limited understanding of its fundamental structure. Despite recent advances in DNA sequencing, a lack of standardized protocols and common analytical frameworks impedes comparisons among studies, hindering the development of global inferences about microbial life on Earth. Here we present a meta-analysis of microbial community samples collected by hundreds of researchers for the Earth Microbiome Project. Coordinated protocols and new analytical methods, particularly the use of exact sequences instead of clustered operational taxonomic units, enable bacterial and archaeal ribosomal RNA gene sequences to be followed across multiple studies and allow us to explore patterns of diversity at an unprecedented scale. The result is both a reference database giving global context to DNA sequence data and a framework for incorporating data from future studies, fostering increasingly complete characterization of Earth's microbial diversity.

Fluid velocities were measured by laser Doppler velocimetry under conditions of pulsatile flow in a scale model of the human carotid bifurcation. Flow velocity and wall shear stress at five axial and four circumferential positions were compared with intimal plaque thickness at corresponding locations in carotid bifurcations obtained from cadavers. Velocities and wall shear stresses during diastole were similar to those found previously under steady flow conditions, but these quantities oscillated in both magnitude and direction during the systolic phase. At the inner wall of the internal carotid sinus, in the region of the flow divider, wall shear stress was highest (systole = 41 dynes/cm2, diastole = 10 dynes/cm2, mean = 17 dynes/cm2) and remained unidirectional during systole. Intimal thickening in this location was minimal. At the outer wall of the carotid sinus where intimal plaques were thickest, mean shear stress was low (-0.5 dynes/cm2) but the instantaneous shear stress oscillated between -7 and +4 dynes/cm2. Along the side walls of the sinus, intimal plaque thickness was greater than in the region of the flow divider and circumferential oscillations of shear stress were prominent. With all 20 axial and circumferential measurement locations considered, strong correlations were found between intimal thickness and the reciprocal of maximum shear stress (r = 0.90, p less than 0.0005) or the reciprocal of mean shear stress (r = 0.82, p less than 0.001). An index which takes into account oscillations of wall shear also correlated strongly with intimal thickness (r = 0.82, p less than 0.001). When only the inner wall and outer wall positions were taken into account, correlations of lesion thickness with the inverse of maximum wall shear and mean wall shear were 0.94 (p less than 0.001) and 0.95 (p less than 0.001), respectively, and with the oscillatory shear index, 0.93 (p less than 0.001). These studies confirm earlier findings under steady flow conditions that plaques tend to form in areas of low, rather than high, shear stress, but indicate in addition that marked oscillations in the direction of wall shear may enhance atherogenesis.

Nickel-Based Superalloys for Advanced Turbine Engines: Chemistry, Microstructure and Properties

The chemical, physical, and mechanical characteristics of nickel-based superalloys are reviewed with emphasis on the use of this class of materials within turbine engines. The role of major and minor alloying additions in multicomponent commercial cast and wrought superalloys is discussed. Microstructural stability and phases observed during processing and in subsequent elevated-temperature service are summarized. Processing paths and recent advances in processing are addressed. Mechanical properties and deformation mechanisms are reviewed, including tensile properties, creep, fatigue, and cyclic crack growth. I.

Combining effect size estimates in meta-analysis with repeated measures and independent-groups designs.

When a meta-analysis on results from experimental studies is conducted, differences in the study design must be taken into consideration. A method for combining results across independent-groups and repeated measures designs is described, and the conditions under which such an analysis is appropriate are discussed. Combining results across designs requires that (a) all effect sizes be transformed into a common metric, (b) effect sizes from each design estimate the same treatment effect, and (c) meta-analysis procedures use design-specific estimates of sampling variance to reflect the precision of the effect size estimates.

Observation of Electron-Antineutrino Disappearance at Daya Bay

The Daya Bay Reactor Neutrino Experiment has measured a nonzero value for the neutrino mixing angle ${\ensuremath{\theta}}_{13}$ with a significance of 5.2 standard deviations. Antineutrinos from six 2.9 $\mathrm{G}{\mathrm{W}}_{\mathrm{th}}$ reactors were detected in six antineutrino detectors deployed in two near (flux-weighted baseline 470 m and 576 m) and one far (1648 m) underground experimental halls. With a $43\text{ }000\text{ }\text{ }\mathrm{ton}--\mathrm{G}{\mathrm{W}}_{\mathrm{th}}--\mathrm{day}$ live-time exposure in 55 days, 10 416 (80 376) electron-antineutrino candidates were detected at the far hall (near halls). The ratio of the observed to expected number of antineutrinos at the far hall is $R=0.940\ifmmode\pm\else\textpm\fi{}\phantom{\rule{0ex}{0ex}}0.011(\mathrm{stat}.)\ifmmode\pm\else\textpm\fi{}0.004(\mathrm{syst}.)$. A rate-only analysis finds ${sin}^{2}2{\ensuremath{\theta}}_{13}=0.092\ifmmode\pm\else\textpm\fi{}0.016(\mathrm{stat}.)\ifmmode\pm\else\textpm\fi{}0.005(\mathrm{syst}.)$ in a three-neutrino framework.

Estimating Effect Sizes From Pretest-Posttest-Control Group Designs

Previous research has recommended several measures of effect size for studies with repeated measurements in both treatment and control groups. Three alternate effect size estimates were compared in terms of bias, precision, and robustness to heterogeneity of variance. The results favored an effect size based on the mean pre-post change in the treatment group minus the mean pre-post change in the control group, divided by the pooled pretest standard deviation.

Abstract Helping students develop informed views of nature of science (NOS) has been and continues to be a central goal for kindergarten through Grade 12 (K–12) science education. Since the early 1960s, major efforts have been undertaken to enhance K–12 students and science teachers' NOS views. However, the crucial component of assessing learners' NOS views remains an issue in research on NOS. This article aims to (a) trace the development of a new open‐ended instrument, the Views of Nature of Science Questionnaire (VNOS), which in conjunction with individual interviews aims to provide meaningful assessments of learners' NOS views; (b) outline the NOS framework that underlies the development of the VNOS; (c) present evidence regarding the validity of the VNOS; (d) elucidate the use of the VNOS and associated interviews, and the range of NOS aspects that it aims to assess; and (e) discuss the usefulness of rich descriptive NOS profiles that the VNOS provides in research related to teaching and learning about NOS. The VNOS comes in response to some calls within the science education community to go back to developing standardized forced‐choice paper and pencil NOS assessment instruments designed for mass administrations to large samples. We believe that these calls ignore much of what was learned from research on teaching and learning about NOS over the past 30 years. The present state of this line of research necessitates a focus on individual classroom interventions aimed at enhancing learners' NOS views, rather than on mass assessments aimed at describing or evaluating students' beliefs. © 2002 Wiley Periodicals, Inc. A bubbling fluidization model using kinetic theory of granular flow

Abstract Detailed knowledge of solids circulation, bubble motion, and frequencies of porosity oscillations is needed for a better understanding of tube erosion in fluidized bed combustors. A predictive two‐phase flow model was derived starting with the Boltzman equation for velocity distribution of particles. The model is a generalization of the Navier‐Stokes equations of the type proposed by R. Jackson, except that the solids viscosities and stresses are computed by simultaneously solving a fluctuating energy equation for the particulate phase. The model predictions agree with time‐averaged and instantaneous porosities measured in two‐dimensional fluidized beds. Observed flow patterns and bubbles were also predicted.

Battery, Ultracapacitor, Fuel Cell, and Hybrid Energy Storage Systems for Electric, Hybrid Electric, Fuel Cell, and Plug-In Hybrid Electric Vehicles: State of the Art

The fuel economy and all-electric range (AER) of hybrid electric vehicles (HEVs) are highly dependent on the onboard energy-storage system (ESS) of the vehicle. Energy-storage devices charge during low power demands and discharge during high power demands, acting as catalysts to provide energy boost. Batteries are the primary energy-storage devices in ground vehicles. Increasing the AER of vehicles by 15% almost doubles the incremental cost of the ESS. This is due to the fact that the ESS of HEVs requires higher peak power while preserving high energy density. Ultracapacitors (UCs) are the options with higher power densities in comparison with batteries. A hybrid ESS composed of batteries, UCs, and/or fuel cells (FCs) could be a more appropriate option for advanced hybrid vehicular ESSs. This paper presents state-of-the-art energy-storage topologies for HEVs and plug-in HEVs (PHEVs). Battery, UC, and FC technologies are discussed and compared in this paper. In addition, various hybrid ESSs that combine two or more storage devices are addressed.

Challenging the Public Stigma of Mental Illness: A Meta-Analysis of Outcome Studies

OBJECTIVE: Public stigma and discrimination have pernicious effects on the lives of people with serious mental illnesses. Given a plethora of research on changing the stigma of mental illness, this article reports on a meta-analysis that examined the effects of antistigma approaches that included protest or social activism, education of the public, and contact with persons with mental illness. METHODS: The investigators heeded published guidelines for systematic literature reviews in health care. This comprehensive and systematic review included articles in languages other than English, dissertations, and population studies. The search included all articles from the inception of the databases until October 2010. Search terms fell into three categories: stigma, mental illness (such as schizophrenia and depression), and change program (including contact and education). The search yielded 72 articles and reports meeting the inclusion criteria of relevance to changing public stigma and sufficient data and statistics to complete analyses. Studies represented 38,364 research participants from 14 countries. Effect sizes were computed for all studies and for each treatment condition within studies. Comparisons between effect sizes were conducted with a weighted one-way analysis of variance. RESULTS: Overall, both education and contact had positive effects on reducing stigma for adults and adolescents with a mental illness. However, contact was better than education at reducing stigma for adults. For adolescents, the opposite pattern was found: education was more effective. Overall, face-to-face contact was more effective than contact by video. CONCLUSIONS: Future research is needed to identify moderators of the effects of both education and contact.

Mental illness stigma: Concepts, consequences, and initiatives to reduce stigma

Persons with mental illness frequently encounter public stigma and may suffer from self-stigma. This review aims to clarify the concept of mental illness stigma and discuss consequences for individuals with mental illness. After a conceptual overview of stigma we discuss two leading concepts of mental illness stigma and consequences of stigma, focussing on self-stigma/empowerment and fear of stigma as a barrier to using health services. Finally, we discuss three main strategies to reduce stigma -- protest, education, and contact -- and give examples of current anti-stigma campaigns. Well-designed anti-stigma initiatives will help to diminish negative consequences of mental illness stigma.

Achieving Secure, Scalable, and Fine-grained Data Access Control in Cloud Computing

Cloud computing is an emerging computing paradigm in which resources of the computing infrastructure are provided as services over the Internet. As promising as it is, this paradigm also brings forth many new challenges for data security and access control when users outsource sensitive data for sharing on cloud servers, which are not within the same trusted domain as data owners. To keep sensitive user data confidential against untrusted servers, existing solutions usually apply cryptographic methods by disclosing data decryption keys only to authorized users. However, in doing so, these solutions inevitably introduce a heavy computation overhead on the data owner for key distribution and data management when fine-grained data access control is desired, and thus do not scale well. The problem of simultaneously achieving fine-grainedness, scalability, and data confidentiality of access control actually still remains unresolved. This paper addresses this challenging open issue by, on one hand, defining and enforcing access policies based on data attributes, and, on the other hand, allowing the data owner to delegate most of the computation tasks involved in fine-grained data access control to untrusted cloud servers without disclosing the underlying data contents. We achieve this goal by exploiting and uniquely combining techniques of attribute-based encryption (ABE), proxy re-encryption, and lazy re-encryption. Our proposed scheme also has salient properties of user access privilege confidentiality and user secret key accountability. Extensive analysis shows that our proposed scheme is highly efficient and provably secure under existing security models.

The Impact of Mental Illness Stigma on Seeking and Participating in Mental Health Care

Treatments have been developed and tested to successfully reduce the symptoms and disabilities of many mental illnesses. Unfortunately, people distressed by these illnesses often do not seek out services or choose to fully engage in them. One factor that impedes care seeking and undermines the service system is mental illness stigma. In this article, we review the complex elements of stigma in order to understand its impact on participating in care. We then summarize public policy considerations in seeking to tackle stigma in order to improve treatment engagement. Stigma is a complex construct that includes public, self, and structural components. It directly affects people with mental illness, as well as their support system, provider network, and community resources. The effects of stigma are moderated by knowledge of mental illness and cultural relevance. Understanding stigma is central to reducing its negative impact on care seeking and treatment engagement. Separate strategies have evolved for counteracting the effects of public, self, and structural stigma. Programs for mental health providers may be especially fruitful for promoting care engagement. Mental health literacy, cultural competence, and family engagement campaigns also mitigate stigma's adverse impact on care seeking. Policy change is essential to overcome the structural stigma that undermines government agendas meant to promote mental health care. Implications for expanding the research program on the connection between stigma and care seeking are discussed.

Limbic-predominant age-related TDP-43 encephalopathy (LATE): consensus working group report

We describe a recently recognized disease entity, limbic-predominant age-related TDP-43 encephalopathy (LATE). LATE neuropathological change (LATE-NC) is defined by a stereotypical TDP-43 proteinopathy in older adults, with or without coexisting hippocampal sclerosis pathology. LATE-NC is a common TDP-43 proteinopathy, associated with an amnestic dementia syndrome that mimicked Alzheimer's-type dementia in retrospective autopsy studies. LATE is distinguished from frontotemporal lobar degeneration with TDP-43 pathology based on its epidemiology (LATE generally affects older subjects), and relatively restricted neuroanatomical distribution of TDP-43 proteinopathy. In community-based autopsy cohorts, ∼25% of brains had sufficient burden of LATE-NC to be associated with discernible cognitive impairment. Many subjects with LATE-NC have comorbid brain pathologies, often including amyloid-β plaques and tauopathy. Given that the 'oldest-old' are at greatest risk for LATE-NC, and subjects of advanced age constitute a rapidly growing demographic group in many countries, LATE has an expanding but under-recognized impact on public health. For these reasons, a working group was convened to develop diagnostic criteria for LATE, aiming both to stimulate research and to promote awareness of this pathway to dementia. We report consensus-based recommendations including guidelines for diagnosis and staging of LATE-NC. For routine autopsy workup of LATE-NC, an anatomically-based preliminary staging scheme is proposed with TDP-43 immunohistochemistry on tissue from three brain areas, reflecting a hierarchical pattern of brain involvement: amygdala, hippocampus, and middle frontal gyrus. LATE-NC appears to affect the medial temporal lobe structures preferentially, but other areas also are impacted. Neuroimaging studies demonstrated that subjects with LATE-NC also had atrophy in the medial temporal lobes, frontal cortex, and other brain regions. Genetic studies have thus far indicated five genes with risk alleles for LATE-NC: GRN, TMEM106B, ABCC9, KCNMB2, and APOE. The discovery of these genetic risk variants indicate that LATE shares pathogenetic mechanisms with both frontotemporal lobar degeneration and Alzheimer's disease, but also suggests disease-specific underlying mechanisms. Large gaps remain in our understanding of LATE. For advances in prevention, diagnosis, and treatment, there is an urgent need for research focused on LATE, including in vitro and animal models. An obstacle to clinical progress is lack of diagnostic tools, such as biofluid or neuroimaging biomarkers, for ante-mortem detection of LATE. Development of a disease biomarker would augment observational studies seeking to further define the risk factors, natural history, and clinical features of LATE, as well as eventual subject recruitment for targeted therapies in clinical trials.

Enabling Public Auditability and Data Dynamics for Storage Security in Cloud Computing

Cloud Computing has been envisioned as the next-generation architecture of IT Enterprise. It moves the application software and databases to the centralized large data centers, where the management of the data and services may not be fully trustworthy. This unique paradigm brings about many new security challenges, which have not been well understood. This work studies the problem of ensuring the integrity of data storage in Cloud Computing. In particular, we consider the task of allowing a third party auditor (TPA), on behalf of the cloud client, to verify the integrity of the dynamic data stored in the cloud. The introduction of TPA eliminates the involvement of the client through the auditing of whether his data stored in the cloud are indeed intact, which can be important in achieving economies of scale for Cloud Computing. The support for data dynamics via the most general forms of data operation, such as block modification, insertion, and deletion, is also a significant step toward practicality, since services in Cloud Computing are not limited to archive or backup data only. While prior works on ensuring remote data integrity often lacks the support of either public auditability or dynamic data operations, this paper achieves both. We first identify the difficulties and potential security problems of direct extensions with fully dynamic data updates from prior works and then show how to construct an elegant verification scheme for the seamless integration of these two salient features in our protocol design. In particular, to achieve efficient data dynamics, we improve the existing proof of storage models by manipulating the classic Merkle Hash Tree construction for block tag authentication. To support efficient handling of multiple auditing tasks, we further explore the technique of bilinear aggregate signature to extend our main result into a multiuser setting, where TPA can perform multiple auditing tasks simultaneously. Extensive security and performance analysis show that the proposed schemes are highly efficient and provably secure.

Power Electronics and Motor Drives in Electric, Hybrid Electric, and Plug-In Hybrid Electric Vehicles

With the requirements for reducing emissions and improving fuel economy, automotive companies are developing electric, hybrid electric, and plug-in hybrid electric vehicles. Power electronics is an enabling technology for the development of these environmentally friendlier vehicles and implementing the advanced electrical architectures to meet the demands for increased electric loads. In this paper, a brief review of the current trends and future vehicle strategies and the function of power electronic subsystems are described. The requirements of power electronic components and electric motor drives for the successful development of these vehicles are also presented.

Privacy-Preserving Public Auditing for Data Storage Security in Cloud Computing

Cloud Computing is the long dreamed vision of computing as a utility, where users can remotely store their data into the cloud so as to enjoy the on-demand high quality applications and services from a shared pool of configurable computing resources. By data outsourcing, users can be relieved from the burden of local data storage and maintenance. However, the fact that users no longer have physical possession of the possibly large size of outsourced data makes the data integrity protection in Cloud Computing a very challenging and potentially formidable task, especially for users with constrained computing resources and capabilities. Thus, enabling public auditability for cloud data storage security is of critical importance so that users can resort to an external audit party to check the integrity of outsourced data when needed. To securely introduce an effective third party auditor (TPA), the following two fundamental requirements have to be met: 1) TPA should be able to efficiently audit the cloud data storage without demanding the local copy of data, and introduce no additional on-line burden to the cloud user; 2) The third party auditing process should bring in no new vulnerabilities towards user data privacy. In this paper, we utilize and uniquely combine the public key based homomorphic authenticator with random masking to achieve the privacy-preserving public cloud data auditing system, which meets all above requirements. To support efficient handling of multiple auditing tasks, we further explore the technique of bilinear aggregate signature to extend our main result into a multi-user setting, where TPA can perform multiple auditing tasks simultaneously. Extensive security and performance analysis shows the proposed schemes are provably secure and highly efficient.

The Double Chooz experiment presents an indication of reactor electron antineutrino disappearance consistent with neutrino oscillations. An observed-to-predicted ratio of events of $0.944\ifmmode\pm\else\textpm\fi{}0.016(\mathrm{stat})\ifmmode\pm\else\textpm\fi{}0.040(\mathrm{syst})$ was obtained in 101 days of running at the Chooz nuclear power plant in France, with two $4.25\text{ }\text{ }{\mathrm{GW}}_{\mathrm{th}}$ reactors. The results were obtained from a single $10\text{ }\text{ }{\mathrm{m}}^{3}$ fiducial volume detector located 1050 m from the two reactor cores. The reactor antineutrino flux prediction used the Bugey4 flux measurement after correction for differences in core composition. The deficit can be interpreted as an indication of a nonzero value of the still unmeasured neutrino mixing parameter ${sin}^{2}2{\ensuremath{\theta}}_{13}$. Analyzing both the rate of the prompt positrons and their energy spectrum, we find ${sin}^{2}2{\ensuremath{\theta}}_{13}=0.086\ifmmode\pm\else\textpm\fi{}0.041(\mathrm{stat})\ifmmode\pm\else\textpm\fi{}0.030(\mathrm{syst})$, or, at 90% C.L., $0.017&lt;{sin}^{2}2{\ensuremath{\theta}}_{13}&lt;0.16$.

On the Self-Stigma of Mental Illness: Stages, Disclosure, and Strategies for Change

People with mental illness have long experienced prejudice and discrimination. Researchers have been able to study this phenomenon as stigma and have begun to examine ways of reducing this stigma. Public stigma is the most prominent form observed and studied, as it represents the prejudice and discrimination directed at a group by the larger population. Self-stigma occurs when people internalize these public attitudes and suffer numerous negative consequences as a result. In our article, we more fully define the concept of self-stigma and describe the negative consequences of self-stigma for people with mental illness. We also examine the advantages and disadvantages of disclosure in reducing the impact of stigma. In addition, we argue that a key to challenging self-stigma is to promote personal empowerment. Lastly, we discuss individual- and societal-level methods for reducing self-stigma, programs led by peers as well as those led by social service providers.
