Natural products in drug discovery: advances and opportunities

Natural products and their structural analogues have historically made a major contribution to pharmacotherapy, especially for cancer and infectious diseases. Nevertheless, natural products also present challenges for drug discovery, such as technical barriers to screening, isolation, characterization and optimization, which contributed to a decline in their pursuit by the pharmaceutical industry from the 1990s onwards. In recent years, several technological and scientific developments - including improved analytical tools, genome mining and engineering strategies, and microbial culturing advances - are addressing such challenges and opening up new opportunities. Consequently, interest in natural products as drug leads is being revitalized, particularly for tackling antimicrobial resistance. Here, we summarize recent technological developments that are enabling natural product-based drug discovery, highlight selected applications and discuss key opportunities.

A critical review on the treatment of dye-containing wastewater: Ecotoxicological and health concerns of textile dyes and possible remediation approaches for environmental safety

The synthetic dyes used in the textile industry pollute a large amount of water. Textile dyes do not bind tightly to the fabric and are discharged as effluent into the aquatic environment. As a result, the continuous discharge of wastewater from a large number of textile industries without prior treatment has significant negative consequences on the environment and human health. Textile dyes contaminate aquatic habitats and have the potential to be toxic to aquatic organisms, which may enter the food chain. This review will discuss the effects of textile dyes on water bodies, aquatic flora, and human health. Textile dyes degrade the esthetic quality of bodies of water by increasing biochemical and chemical oxygen demand, impairing photosynthesis, inhibiting plant growth, entering the food chain, providing recalcitrance and bioaccumulation, and potentially promoting toxicity, mutagenicity, and carcinogenicity. Therefore, dye-containing wastewater should be effectively treated using eco-friendly technologies to avoid negative effects on the environment, human health, and natural water resources. This review compares the most recent technologies which are commonly used to remove dye from textile wastewater, with a focus on the advantages and drawbacks of these various approaches. This review is expected to spark great interest among the research community who wish to combat the widespread risk of toxic organic pollutants generated by the textile industries.

Metal‐Assisted Chemical Etching of Silicon: A Review

This article presents an overview of the essential aspects in the fabrication of silicon and some silicon/germanium nanostructures by metal-assisted chemical etching. First, the basic process and mechanism of metal-assisted chemical etching is introduced. Then, the various influences of the noble metal, the etchant, temperature, illumination, and intrinsic properties of the silicon substrate (e.g., orientation, doping type, doping level) are presented. The anisotropic and the isotropic etching behaviors of silicon under various conditions are presented. Template-based metal-assisted chemical etching methods are introduced, including templates based on nanosphere lithography, anodic aluminum oxide masks, interference lithography, and block-copolymer masks. The metal-assisted chemical etching of other semiconductors is also introduced. A brief introduction to the application of Si nanostructures obtained by metal-assisted chemical etching is given, demonstrating the promising potential applications of metal-assisted chemical etching. Finally, some open questions in the understanding of metal-assisted chemical etching are compiled.

Changing cancer survival in China during 2003–15: a pooled analysis of 17 population-based cancer registries

BACKGROUND: From 2003 to 2005, standardised 5-year cancer survival in China was much lower than in developed countries and varied substantially by geographical area. Monitoring population-level cancer survival is crucial to the understanding of the overall effectiveness of cancer care. We therefore aimed to investigate survival statistics for people with cancer in China between 2003 and 2015. METHODS: We used population-based data from 17 cancer registries in China. Data for the study population was submitted by the end of July 31, 2016, with follow-up data on vital status obtained on Dec 31, 2015. We used anonymised, individual cancer registration records of patients (aged 0-99 years) diagnosed with primary, invasive cancers from 2003 to 2013. Patients eligible for inclusion had data for demographic characteristics, date of diagnosis, anatomical site, morphology, behaviour code, vital status, and last date of contact. We analysed 5-year relative survival by sex, age, and geographical area, for all cancers combined and 26 different cancer types, between 2003 and 2015. We stratified survival estimates by calendar period (2003-05, 2006-08, 2009-11, and 2012-15). FINDINGS: There were 678 842 records of patients with invasive cancer who were diagnosed between 2003 and 2013. Of these records, 659 732 (97·2%) were eligible for inclusion in the final analyses. From 2003-05 to 2012-15, age-standardised 5-year relative survival increased substantially for all cancers combined, for both male and female patients, from 30·9% (95% CI 30·6-31·2) to 40·5% (40·3-40·7). Age-standardised 5-year relative survival also increased for most cancer types, including cancers of the uterus (average change per calendar period 5·5% [95% CI 2·5-8·5]), thyroid (5·4% [3·2-7·6]), cervix (4·5% [2·9-6·2]), and bone (3·2% [2·1-4·4]). In 2012-15, age-standardised 5-year survival for all patients with cancer was higher in urban areas (46·7%, 95% CI 46·5-47·0) than in rural areas (33·6%, 33·3-33·9), except for patients with oesophageal or cervical cancer; but improvements in survival were greater for patients residing in rural areas than in urban areas. Relative survival decreased with increasing age. The increasing trends in survival were consistent with the upward trends of medical expenditure of the country during the period studied. INTERPRETATION: There was a marked overall increase in cancer survival from 2003 to 2015 in the population covered by these cancer registries in China, possibly reflecting advances in the quality of cancer care in these areas. The survival gap between urban and rural areas narrowed over time, although geographical differences in cancer survival remained. Insight into these trends will help prioritise areas that need increased cancer care. Pyroptosis: A new frontier in cancer

Pyroptosis is an inflammatory form of cell death triggered by certain inflammasomes, leading to the cleavage of gasdermin D (GSDMD) and activation of inactive cytokines like IL-18 and IL-1β. Pyroptosis has been reported to be closely associated to some diseases like atherosclerosis and diabetic nephropathy. Recently, some studies found that pyroptosis can influence the proliferation, invasion and metastasis of tumor, which regulated by some non-coding RNAs and other molecules. Hence, we provided an overview of morphological and molecular characteristics of pyroptosis. We also focus on mechanism of regulating pyroptosis in tumor cells as well as the potential roles of pyroptosis in cancer to explore potential diagnostic markers in cancers contributing to the prevention and treatment in cancers.

Characterization of Surface EMG Signal Based on Fuzzy Entropy

Fuzzy entropy (FuzzyEn), a new measure of time series regularity, was proposed and applied to the characterization of surface electromyography (EMG) signals. Similar to the two existing related measures ApEn and SampEn, FuzzyEn is the negative natural logarithm of the conditional probability that two vectors similar for m points remain similar for the next m + 1 points. Importing the concept of fuzzy sets, vectors' similarity is fuzzily defined in FuzzyEn on the basis of exponential function and their shapes. Besides possessing the good properties of SampEn superior to ApEn, FuzzyEn also succeeds in giving the entropy definition in the case of small parameters. Its performance on characterizing surface EMG signals, as well as independent, identically distributed (i.i.d.) random numbers and periodical sinusoidal signals, shows that FuzzyEn can more efficiently measure the regularity of time series. The method introduced here can also be applied to other noisy physiological signals with relatively short datasets.

A Hierarchical Z‑Scheme α‐Fe<sub>2</sub>O<sub>3</sub>/g‐C<sub>3</sub>N<sub>4</sub> Hybrid for Enhanced Photocatalytic CO<sub>2</sub> Reduction

Abstract The challenge in the artificial photosynthesis of fossil resources from CO 2 by utilizing solar energy is to achieve stable photocatalysts with effective CO 2 adsorption capacity and high charge‐separation efficiency. A hierarchical direct Z‐scheme system consisting of urchin‐like hematite and carbon nitride provides an enhanced photocatalytic activity of reduction of CO 2 to CO, yielding a CO evolution rate of 27.2 µmol g −1 h −1 without cocatalyst and sacrifice reagent, which is &gt;2.2 times higher than that produced by g‐C 3 N 4 alone (10.3 µmol g −1 h −1 ). The enhanced photocatalytic activity of the Z‐scheme hybrid material can be ascribed to its unique characteristics to accelerate the reduction process, including: (i) 3D hierarchical structure of urchin‐like hematite and preferable basic sites which promotes the CO 2 adsorption, and (ii) the unique Z‐scheme feature efficiently promotes the separation of the electron–hole pairs and enhances the reducibility of electrons in the conduction band of the g‐C 3 N 4 . The origin of such an obvious advantage of the hierarchical Z‐scheme is not only explained based on the experimental data but also investigated by modeling CO 2 adsorption and CO adsorption on the three different atomic‐scale surfaces via density functional theory calculation. Micro/Nanoparticle‐Augmented Sonodynamic Therapy (SDT): Breaking the Depth Shallow of Photoactivation

The fast development of photoactivation for cancer treatment provides an efficient photo-therapeutic strategy for cancer treatment, but traditional photodynamic or photothermal therapy suffers from the critical issue of low in vivo penetration depth of tissues. As a non-invasive therapeutic modality, sonodynamic therapy (SDT) can break the depth barrier of photoactivation because ultrasound has an intrinsically high tissue-penetration performance. Micro/nanoparticles can efficiently augment the SDT efficiency based on nanobiotechnology. The state-of-art of the representative achievements on micro/nanoparticle-enhanced SDT is summarized, and specific functions of micro/nanoparticles for SDT are discussed, from the different viewpoints of ultrasound medicine, material science and nanobiotechnology. Emphasis is put on the relationship of structure/composition-SDT performance of micro/nanoparticle-based sonosensitizers. Three types of micro/nanoparticle-augmented SDT are discussed, including organic and inorganic sonosensitizers and micro/nanoparticle-based but sonosensitizer-free strategies to enhance the SDT outcome. SDT-based synergistic cancer therapy augmented by micro/nanoparticles and their biosafety are also included. Some urgent critical issues and potential developments of micro/nanoparticle-augmented SDT for efficient cancer treatment are addressed. It is highly expected that micro/nanoparticle-augmented SDT will be quickly developed as a new and efficient therapeutic modality which will find practical applications in cancer treatment. At the same time, fundamental disciplines regarding materials science, chemistry, medicine and nanotechnology will be advanced.

Exosomes Derived from Human Umbilical Cord Mesenchymal Stem Cells Alleviate Liver Fibrosis

Mesenchymal stem cells (MSCs) have been considered as an attractive tool for the therapy of diseases. Exosomes excreted from MSCs can reduce myocardial ischemia/reperfusion damage and protect against acute tubular injury. However, whether MSC-derived exosomes can relieve liver fibrosis and its mechanism remain unknown. Previous work showed that human umbilical cord-MSCs (hucMSCs) transplanted into acutely injured and fibrotic livers could restore liver function and improve liver fibrosis. In this study, it was found that transplantation of exosomes derived from hucMSC (hucMSC-Ex) reduced the surface fibrous capsules and got their textures soft, alleviated hepatic inflammation and collagen deposition in carbon tetrachloride (CCl4)-induced fibrotic liver. hucMSC-Ex also significantly recovered serum aspartate aminotransferase (AST) activity, decreased collagen type I and III, transforming growth factor (TGF)-β1 and phosphorylation Smad2 expression in vivo. In further experiments, we found that epithelial-to-mesenchymal transition (EMT)-associated markers E-cadherin-positive cells increased and N-cadherin- and vimentin-positive cells decreased after hucMSC-Ex transplantation. Furthermore, the human liver cell line HL7702 underwent typical EMT after induction with recombinant human TGF-β1, and then hucMSC-Ex treatment reversed spindle-shaped and EMT-associated markers expression in vitro. Taken together, these results suggest that hucMSC-Ex could ameliorate CCl4-induced liver fibrosis by inhibiting EMT and protecting hepatocytes. This provides a novel approach for the treatment of fibrotic liver disease.

Exosomes as a new frontier of cancer liquid biopsy

Liquid biopsy, characterized by minimally invasive detection through biofluids such as blood, saliva, and urine, has emerged as a revolutionary strategy for cancer diagnosis and prognosis prediction. Exosomes are a subset of extracellular vesicles (EVs) that shuttle molecular cargoes from donor cells to recipient cells and play a crucial role in mediating intercellular communication. Increasing studies suggest that exosomes have a great promise to serve as novel biomarkers in liquid biopsy, since large quantities of exosomes are enriched in body fluids and are involved in numerous physiological and pathological processes. However, the further clinical application of exosomes has been greatly restrained by the lack of high-quality separation and component analysis methods. This review aims to provide a comprehensive overview on the conventional and novel technologies for exosome isolation, characterization and content detection. Additionally, the roles of exosomes serving as potential biomarkers in liquid biopsy for the diagnosis, treatment monitoring, and prognosis prediction of cancer are summarized. Finally, the prospects and challenges of applying exosome-based liquid biopsy to precision medicine are evaluated.

Lattice oxygen activation enabled by high-valence metal sites for enhanced water oxidation

Abstract Anodic oxygen evolution reaction (OER) is recognized as kinetic bottleneck in water electrolysis. Transition metal sites with high valence states can accelerate the reaction kinetics to offer highly intrinsic activity, but suffer from thermodynamic formation barrier. Here, we show subtle engineering of highly oxidized Ni 4+ species in surface reconstructed (oxy)hydroxides on multicomponent FeCoCrNi alloy film through interatomically electronic interplay. Our spectroscopic investigations with theoretical studies uncover that Fe component enables the formation of Ni 4+ species, which is energetically favored by the multistep evolution of Ni 2+ →Ni 3+ →Ni 4+ . The dynamically constructed Ni 4+ species drives holes into oxygen ligands to facilitate intramolecular oxygen coupling, triggering lattice oxygen activation to form Fe-Ni dual-sites as ultimate catalytic center with highly intrinsic activity. As a result, the surface reconstructed FeCoCrNi OER catalyst delivers outstanding mass activity and turnover frequency of 3601 A g metal −1 and 0.483 s −1 at an overpotential of 300 mV in alkaline electrolyte, respectively.

Overview of Stator-Permanent Magnet Brushless Machines

Permanent magnet (PM) brushless machines having magnets and windings in stator (the so-called stator-PM machines) have attracted more and more attention in the past decade due to its definite advantages of robust structure, high power density, high efficiency, etc. In this paper, an overview of the stator-PM machine is presented, with particular emphasis on concepts, operation principles, machine topologies, electromagnetic performance, and control strategies. Both brushless ac and dc operation modes are described. The key features of the machines, including the merits and drawbacks of the machines, are summarized. Moreover, the latest development of the machines is also discussed.

HucMSC-Exosome Mediated-Wnt4 Signaling Is Required for Cutaneous Wound Healing

Mesenchymal stem cell-derived exosomes (MSC-Ex) play important roles in tissue injury repair, however, the roles of MSC-Ex in skin damage repair and its mechanisms are largely unknown. Herein, we examined the benefit of human umbilical cord MSC-derived exosome (hucMSC-Ex) in cutaneous wound healing using a rat skin burn model. We found that hucMSC-Ex-treated wounds exhibited significantly accelerated re-epithelialization, with increased expression of CK19, PCNA, collagen I (compared to collagen III) in vivo. HucMSC-Ex promoted proliferation and inhibited apoptosis of skin cells after heat-stress in vitro. We also discovered that Wnt4 was contained in hucMSC-Ex, and hucMSC-Ex-derived Wnt4 promoted β-catenin nuclear translocation and activity to enhance proliferation and migration of skin cells, which could be reversed by β-catenin inhibitor ICG001. In vivo studies confirmed that the activation of Wnt/β-catenin by hucMSC-Ex played a key role in wound re-epithelialization and cell proliferation. Furthermore, knockdown of Wnt4 in hucMSC-Ex abrogated β-catenin activation and skin cell proliferation and migration in vitro. The in vivo therapeutic effects were also inhibited when the expression of Wnt4 in hucMSC-Ex was interfered. In addition, the activation of AKT pathway by hucMSC-Ex was associated with the reduction of heat stress-induced apoptosis in rat skin burn model. Collectively, our findings indicate that exosome-delivered Wnt4 provides new aspects for the therapeutic strategy of MSCs in cutaneous wound healing. Stem Cells 2015;33:2158-2168.

High Efficiency Photocatalytic Water Splitting Using 2D α‐Fe<sub>2</sub>O<sub>3</sub>/g‐C<sub>3</sub>N<sub>4</sub> Z‐Scheme Catalysts

Photocatalysis is the most promising method for achieving artificial photosynthesis, but a bottleneck is encountered in finding materials that could efficiently promote the water splitting reaction. The nontoxicity, low cost, and versatility of photocatalysts make them especially attractive for this application. This study demonstrates that small amounts of α‐Fe 2 O 3 nanosheets can actively promote exfoliation of g‐C 3 N 4 , producing 2D hybrid that exhibits tight interfaces and an all‐solid‐state Z‐scheme junction. These nanostructured hybrids present a high H 2 evolution rate &gt;3 × 10 4 µmol g ‐1 h ‐1 and external quantum efficiency of 44.35% at λ = 420 nm, the highest value so far reported among the family of g‐C 3 N 4 photocatalysts. Besides effectively suppressing the recombination of electron–hole pairs, this Z‐scheme junction also exhibits activity toward overall water splitting without any sacrificial donor. The proposed synthetic route for controlled production of 2D g‐C 3 N 4 ‐based structures provides a scalable alternative toward the development of highly efficient and active photocatalysts.

Exosomes in cancer: small particle, big player

Exosomes have emerged as a novel mode of intercellular communication. Exosomes can shuttle bioactive molecules including proteins, DNA, mRNA, as well as non-coding RNAs from one cell to another, leading to the exchange of genetic information and reprogramming of the recipient cells. Increasing evidence suggests that tumor cells release excessive amount of exosomes, which may influence tumor initiation, growth, progression, metastasis, and drug resistance. In addition, exosomes transfer message from tumor cells to immune cells and stromal cells, contributing to the escape from immune surveillance and the formation of tumor niche. In this review, we highlight the recent advances in the biology of exosomes as cancer communicasomes. We review the multifaceted roles of exosomes, the small secreted particles, in communicating with other cells within tumor microenvironment. Given that exosomes are cell type specific, stable, and accessible from body fluids, exosomes may provide promising biomarkers for cancer diagnosis and represent new targets for cancer therapy.

Maximum Neighborhood Margin Discriminant Projection for Classification

We develop a novel maximum neighborhood margin discriminant projection (MNMDP) technique for dimensionality reduction of high-dimensional data. It utilizes both the local information and class information to model the intraclass and interclass neighborhood scatters. By maximizing the margin between intraclass and interclass neighborhoods of all points, MNMDP cannot only detect the true intrinsic manifold structure of the data but also strengthen the pattern discrimination among different classes. To verify the classification performance of the proposed MNMDP, it is applied to the PolyU HRF and FKP databases, the AR face database, and the UCI Musk database, in comparison with the competing methods such as PCA and LDA. The experimental results demonstrate the effectiveness of our MNMDP in pattern classification.

Microalgae-based wastewater treatment: Mechanisms, challenges, recent advances, and future prospects

The rapid expansion of both the global economy and the human population has led to a shortage of water resources suitable for direct human consumption. As a result, water remediation will inexorably become the primary focus on a global scale. Microalgae can be grown in various types of wastewaters (WW). They have a high potential to remove contaminants from the effluents of industries and urban areas. This review focuses on recent advances on WW remediation through microalgae cultivation. Attention has already been paid to microalgae-based wastewater treatment (WWT) due to its low energy requirements, the strong ability of microalgae to thrive under diverse environmental conditions, and the potential to transform WW nutrients into high-value compounds. It turned out that microalgae-based WWT is an economical and sustainable solution. Moreover, different types of toxins are removed by microalgae through biosorption, bioaccumulation, and biodegradation processes. Examples are toxins from agricultural runoffs and textile and pharmaceutical industrial effluents. Microalgae have the potential to mitigate carbon dioxide and make use of the micronutrients that are present in the effluents. This review paper highlights the application of microalgae in WW remediation and the remediation of diverse types of pollutants commonly present in WW through different mechanisms, simultaneous resource recovery, and efficient microalgae-based co-culturing systems along with bottlenecks and prospects.

Surface Defect Engineering in 2D Nanomaterials for Photocatalysis

Abstract 2D Nanomaterials, with unique structural and electronic features, have shown enormous potential toward photocatalysis fields. However, the photocatalytic behavior of pristine 2D photocatalysts are still unsatisfactory, and far below the requirements of practical applications. In this regard, surface defect engineering can serve as an effective means to tune photoelectric parameters of 2D photocatalysts through tailoring the local surface microstructure, electronic structure, and carrier concentration. In this review, recent progress in the design of surface defects with the classified anion vacancy, cation vacancy, vacancy associates, pits, distortions, and disorder on 2D photocatalysts to boost the photocatalytic performance is summarized. The strategies for controlling defects formation and technique to distinguish various surface defects are presented. The crucial roles of surface defects for photocatalysis performance optimization are proposed and advancement of defective 2D photocatalysts toward versatile applications such as water oxidation, hydrogen production, CO 2 reduction, nitrogen fixation, organic synthesis, and pollutants removal are discussed. Surface defect modulated 2D photocatalysts thus represent a powerful configuration for further development toward photocatalysis.
