Id1 overexpression is independent of repression and epigenetic silencing of tumor suppressor genes in melanoma

The full molecular consequences of oncogene activation during tumorigenesis are not well understood, but several studies have recently linked oncogene activation to epigenetic silencing of specific genes 1, 2. Transcriptional repressor Id1 is overexpressed in many malignancies including melanoma, and Id1 targets include tumor suppressor genes TSP1, CDKN2A (p16) and CDKN1A (p21), which are frequently epigenetically silenced in cancer. We confirmed that both TSP1 and CDKN2A have abnormal promoter region DNA methylation in primary melanoma, but the mechanism by which this silencing occurs remains unknown. Here we explore the effects of stable lentiviral Id1 overexpression on the expression of these Id1 target genes in human melanoma cell lines. Overexpressed Id1 was functional and bound transcriptional activator E2A, but did not sequester E2A from gene promoters and repress gene expression. Therefore, these Id1 target genes were resistant to Id1-mediated gene silencing. Our results suggest that Id1 activation may need to occur at discrete stages in cooperation with additional gene dysregulation to repress and induce epigenetic silencing of tumor suppressor genes during melanoma progression.

They may impinge on the dignity of the patient as well as subject society to unwarranted expense. In order to provide affordable healthcare for all, these concepts are in need of modification.

Suicide Trends in Jordan in Correlation With the COVID-19 Pandemic: A Forensic Medicine Perspective

Background This study aimed to examine the suicide trends among Jordanians who died by suicide before and during the coronavirus disease 2019 (COVID-19) pandemic and assess their correlation with demographic data, including gender, age, marital status, and the method used for suicide, from a forensic medicine perspective. Methodology This was a retrospective, observational, autopsy-based study. Of the 731 medicolegal autopsies performed at the Al-Hussein New Hospital at Al Salt City between 2017 and 2022, 41 confirmed cases of suicide were identified. Results The study indicated that 75% of the subjects committed suicide during the COVID-19 pandemic period, of whom 13% were children. The average age of those who died by suicide was 32.1 (SD = 12.2) years. Overall, 58.5% were single, 61.0% unemployed, and 53.7% had a mental illness. Males were four times more likely to commit suicide than females. Hanging represented 60.61% of all male suicides, followed by firearms (27.27%); however, females used poisoning. Around 60% and 25% of the study subjects were in the age group of 21-40 years and 41 and more, respectively. The study also identified a statistically significant association between gender, age, and the time of suicide (during COVID-19). The findings of this study could not confirm any association between suicide and mental illness, marital status, and employment status. Conclusions The suicide rate in Jordan increased during the COVID-19 pandemic, and it is alarming that most of those who committed suicide were at a young age and in their productive years.

هدفت الدراسة إلى التعرف على مفهوم خطاب الكراهية، ودور وسائل التواصل الاجتماعي في انتشار خطاب الكراهية من وجهة نظر الصحفيين الأردنيين، ولتحقيق أهداف الدراسة تم الاعتماد على المنهج الوصفي التحليلي، وتصميم أداة للدراسة (استبيان) تم توزيعه على عينة من الصحفيين الأردنيين وبواقع (300) صحفي. وتوصلت الدراسة إلى مجموعة من النتائج من أهمها: أن لمواقع التواصل الاجتماعي دوراً في نشر خطاب الكراهية، وأن انتشار خطاب الكراهية يثير الفتن بين مكونات المجتمع، بالإضافة إلى وجود أثر ذو دلالة إحصائیة لوسائل التواصل الاجتماعي في انتشار خطاب الكراهية من وجهة نظر الصحفيين الأردنيين. وقد أوصت الدراسة بالعمل على تكثیف التوعیة حول مفهوم الكراهیة وخطابات الكراهیة لیقوم الناس بالحذر منها وتجنبها، وأنه على الصحفيين مستخدمي شبكات التواصل الاجتماعي الابتعاد عن نشر المعلومات دون التأكد من صحتها، والابتعاد كذلك عن تشجيع الآراء والأفكار المتطرفة.

Autopsy-Based Study of Non-accidental Violent Neck Asphyxia in Jordan: A Retrospective Study

Background There is a global increase in the number of deaths caused by violent neck asphyxia, which includes both suicides and homicides. This research presents autopsy-based findings and analytical demographic data that are pertinent to significant medicolegal cases. Methods A retrospective examination of 113 autopsy reports of non-accidental violent neck asphyxia reported to the forensic pathology department of Al-Balqa Governorate in Jordan within the period from January 2010 to March 2023. Age, gender, and autopsy results were documented, along with medicolegal records that included toxicology reports. Conversely, information on the manner of death was derived from police inquest records. For the purpose of this research, all the cases of throttling and ligature strangulation have been determined to be homicidal, and all cases of hanging were classified as suicide. Results Males were predominant in hanging (72%), while females were predominant in ligature strangulation (82%). The mean age group in hanging was 21-35 years (51%), while the range for throttling was 36-50 years, and that in strangulation was 2-20 years. Most hanging and throttling cases showed typical external neck findings. Seventy-two percent of hanging cases (n = 68) did not exhibit any internal neck findings, while all throttling cases yielded positive results. Both hanging and strangulation by ligature cases exhibited an absence of thyroid cartilage fracture (83%). Eighty-nine cases (95%) of hanging did not exhibit any injuries to the rest of the body. However, all cases of throttling and ligature strangulation had an almost positive external body injury. In all cases of throttling and ligature strangulation, ocular petechial hemorrhage was present, whereas one-third of the hanging cases had it. Conclusion The forensic doctor must observe the pattern of ligature marks and other signs of physical assault found on the neck, meticulously examine the rest of the body, rule out other causes of death, and cooperate with the legal authorities regarding the collection of the history and death scene findings to accurately determine the manner and mechanism of death in cases of violent neck asphyxia.

Recurrent Indolent Keratitis Caused by Achromobacter xylosoxidans: A Case Report and Literature Review

An 83-year-old male with a history of radial keratotomy and laser-assisted in situ keratomileusis (LASIK) presented with symptoms of a non-resolving corneal ulcer in the right eye that had been present for five months. The patient was treated with antibacterial, antiviral, and antifungal medications over that period, with multiple recurrences that prompted referral to our tertiary center for management. Following a 48-hour cessation of all medications, a corneal biopsy was performed which grew Achromobacter xylosoxidans. In conclusion, Achromobacter xylosoxidans remains a rare cause of keratitis but should be considered in patients with a slow-progressing disease. Risk factors include previous corneal surgeries or trauma, topical steroid use, and contact lens wear. Because A. xylosoxidans grows slowly, it can have a deep impact on the cornea. To help antibiotics get into the cornea properly, epithelial debridement may be helpful. It does not respond to standard empiric antibiotic therapy. Ceftazidime and fourth-generation fluoroquinolones are better choices for treatment.

The Role of the Health Media in Raising Awareness of the Emerging Corona Virus Vaccine: A Field Study from a Point of View on Jordanian Opinion Leaders

study aimed to identify the role of the health media in relation to Corona virus vaccination from the perspective of opinion leaders, and the researcher used a descriptive analytical approach, selecting a random sample of Jordanian opinion leaders (50), distributing a questionnaire to them, and analyzing the results of their responses. The study found that all members of the study sample of opinion leaders received the Corona vaccine, of which (54.0 %) received the Pfizer vaccine, 30.0 % received the AstraZeneca vaccine, and 16.0 % received the Sino-pharma vaccine, and that (88.0 %) prefer Pfizer vaccine, while (10.0 %) prefer Sino-pharm, and (6.0 %) prefer AstraZeneca vaccine. The findings also revealed that (50.0 percent) of the study sample believed that the main reason for people's reluctance to take the Corona vaccine is fear of side effects, and that the majority of the sample members of Jordanian opinion leaders agreed that the information provided by the Ministry of Health in Jordanian media was sufficient to dispel rumors to a large extent, as it clarified the effectiveness and safety of vaccinations. The study recommended the need for official institutions, particularly Jordan TV and the Ministry of Health, and through various media, to continue to provide sufficient information about the coronavirus disease, and its catastrophic consequences on the economic, health and social levels, as well as the need for official authorities to provide through the various media accurate information based on global sources in the positive Receiving the Corona vaccine, its effectiveness in fighting the disease, and its safety for individuals. Elderly Patient With Bilateral Central Serous Chorioretinopathy Misdiagnosed as Bilateral Wet Age-Related Macular Degeneration: A Case Report

This report presents a unique case of a 77-year-old diabetic male patient with bilateral central serous chorioretinopathy (CSCR), who was receiving multiple bilateral intravitreal injections for a presumed diagnosis of wet age-related macular degeneration (AMD). The fundus examination did not show any signs of AMD or diabetic retinopathy (DR). The spectral domain optical coherence tomography (OCT) revealed bilateral subretinal fluid. The neovascular membrane was not visible on OCT angiography. Fundus fluorescein angiography (FFA) confirmed the absence of choroidal neovascularization (CNV). Notably, this represents a unique case of an elderly patient with CSCR mimicking occult CNV.

Quality of Media Training in Jordan

The purpose of this study is twofold. The first is to explore the real aspect of media training in Jordan. The second is to assess the quality of media education and training from a journalist's perspective. In so doing, this study aims at shaping a scientific vision of the reality of training along with the required priorities in an attempt to match the quality standards for media training. This study is based on survey sampling, both descriptive and analytical. A sample of 212 journalists is drawn from all the 1800 Jordanian journalists working in civil, private and public, media institutions. By way of concluding, this study revealed that the level of quality training in Jordan is low from a journalist's perspective. Further, the methods of teaching journalism and media in Jordanian universities are not in line with the standards for quality education and qualification in media sectors. On the side, the problems hindering the development of the quality of media training are the weakness of trainers and trainees, the modesty in spending allocations on training, and poor coordination among the stakeholders of media training. Among other factors is also the absence of national independent institutions of media training. More importantly, this study emphasized that there is a statistical relation between the awareness of journalists of the resources of quality training and the kind of (whether private, contributed, public or governmental) ownership of media outlets
