In a previous clinical study, a probiotic formulation (PF) consisting of Lactobacillus helveticus R0052 and Bifidobacterium longum R0175 (PF) decreased stress-induced gastrointestinal discomfort. Emerging evidence of a role for gut microbiota on central nervous system functions therefore suggests that oral intake of probiotics may have beneficial consequences on mood and psychological distress. The aim of the present study was to investigate the anxiolytic-like activity of PF in rats, and its possible effects on anxiety, depression, stress and coping strategies in healthy human volunteers. In the preclinical study, rats were daily administered PF for 2 weeks and subsequently tested in the conditioned defensive burying test, a screening model for anti-anxiety agents. In the clinical trial, volunteers participated in a double-blind, placebo-controlled, randomised parallel group study with PF administered for 30 d and assessed with the Hopkins Symptom Checklist (HSCL-90), the Hospital Anxiety and Depression Scale (HADS), the Perceived Stress Scale, the Coping Checklist (CCL) and 24 h urinary free cortisol (UFC). Daily subchronic administration of PF significantly reduced anxiety-like behaviour in rats (P < 0·05) and alleviated psychological distress in volunteers, as measured particularly by the HSCL-90 scale (global severity index, P < 0·05; somatisation, P < 0·05; depression, P < 0·05; and anger-hostility, P < 0·05), the HADS (HADS global score, P < 0·05; and HADS-anxiety, P < 0·06), and by the CCL (problem solving, P < 0·05) and the UFC level (P < 0·05). L. helveticus R0052 and B. longum R0175 taken in combination display anxiolytic-like activity in rats and beneficial psychological effects in healthy human volunteers.
Abstract Important operational changes that have gradually been assimilated and new approaches that are developing as part of the movement toward sustainable intensive aquaculture production systems are presented via historical, current, and future perspectives. Improved environmental and economic sustainability based on increased efficiency of production continues to be realized. As a result, aquaculture continues to reduce its carbon footprint through reduced greenhouse gas emissions. Reduced use of freshwater and land resources per unit of production, improved feed management practices as well as increased knowledge of nutrient requirements, effective feed ingredients and additives, domestication of species, and new farming practices are now being applied or evaluated. Successful expansion into culture of marine species, both off and on shore, offers the potential of substantial increases in sustainable intensive aquaculture production combined with integrative efforts to increase efficiency will principally contribute to satisfying the increasing global demand for protein and food security needs.
Alzheimer's disease (AD) is the most frequent case of neurodegenerative disease and is becoming a major public health problem all over the world. Many therapeutic strategies have been explored for several decades; however, there is still no curative treatment, and the priority remains prevention. In this review, we present an update on the clinical and physiological phase of the AD spectrum, modifiable and non-modifiable risk factors for AD treatment with a focus on prevention strategies, then research models used in AD, followed by a discussion of treatment limitations. The prevention methods can significantly slow AD evolution and are currently the best strategy possible before the advanced stages of the disease. Indeed, current drug treatments have only symptomatic effects, and disease-modifying treatments are not yet available. Drug delivery to the central nervous system remains a complex process and represents a challenge for developing therapeutic and preventive strategies. Studies are underway to test new techniques to facilitate the bioavailability of molecules to the brain. After a deep study of the literature, we find the use of soft nanoparticles, in particular nanoliposomes and exosomes, as an innovative approach for preventive and therapeutic strategies in reducing the risk of AD and solving problems of brain bioavailability. Studies show the promising role of nanoliposomes and exosomes as smart drug delivery systems able to penetrate the blood-brain barrier and target brain tissues. Finally, the different drug administration techniques for neurological disorders are discussed. One of the promising therapeutic methods is the intranasal administration strategy which should be used for preclinical and clinical studies of neurodegenerative diseases.
High O(2) consumption, modest antioxidant defenses and a lipid-rich constitution make the brain highly vulnerable to redox imbalances. Oxidative damage in the brain causes nervous system impairment. Recently, oxidative stress has also been implicated in depression, anxiety disorders and high anxiety levels. The findings which establish a link between oxidative stress and pathological anxiety have inspired a number of other recent studies focusing on the link between oxidative status and normal anxiety and also on a possible causal relationship between cellular oxidative stress and emotional stress. This review examines the recent discoveries made on the link between oxidative status and normal anxiety levels and the putative role of oxidative stress in genesis of anxiety. We discuss the different opinions and questions that exist in the field and review the methodological approaches that are being used to determine a causal relationship between oxidative and emotional stress.
The relative bioavailability of arsenic, antimony, cadmium, and lead for the ingestion pathway was measured in 16 soils contaminated by either smelting or mining activities using a juvenile swine model. The soils contained 18 to 25,000 mg kg(-1) As, 18 to 60,000 mg kg(-1) Sb, 20 to 184 mg kg(-1) Cd, and 1460 to 40,214 mg kg(-1) Pb. The bioavailability in the soils was measured in kidney, liver, bone, and urine relative to soluble salts of the four elements. The variety of soil types, the total concentrations of the elements, and the range of bioavailabilities found were considered to be suitable for calibrating the in vitro Unified BARGE bioaccessibility method. The bioaccessibility test has been developed by the BioAccessibility Research Group of Europe (BARGE) and is known as the Unified BARGE Method (UBM). The study looked at four end points from the in vivo measurements and two compartments in the in vitro study ("stomach" and "stomach and intestine"). Using benchmark criteria for assessing the "fitness for purpose" of the UBM bioaccessibility data to act as an analogue for bioavailability in risk assessment, the study shows that the UBM met criteria on repeatability (median relative standard deviation value <10%) and the regression statistics (slope 0.8 to 1.2 and r-square > 0.6) for As, Cd, and Pb. The data suggest a small bias in the UBM relative bioaccessibility of As and Pb compared to the relative bioavailability measurements of 3% and 5% respectively. Sb did not meet the criteria due to the small range of bioaccessibility values found in the samples.
Agroecology and industrial ecology can be viewed as complementary means for reducing the environmental footprint of animal farming systems: agroecology mainly by stimulating natural processes to reduce inputs, and industrial ecology by closing system loops, thereby reducing demand for raw materials, lowering pollution and saving on waste treatment. Surprisingly, animal farming systems have so far been ignored in most agroecological thinking. On the basis of a study by Altieri, who identified the key ecological processes to be optimized, we propose five principles for the design of sustainable animal production systems: (i) adopting management practices aiming to improve animal health, (ii) decreasing the inputs needed for production, (iii) decreasing pollution by optimizing the metabolic functioning of farming systems, (iv) enhancing diversity within animal production systems to strengthen their resilience and (v) preserving biological diversity in agroecosystems by adapting management practices. We then discuss how these different principles combine to generate environmental, social and economic performance in six animal production systems (ruminants, pigs, rabbits and aquaculture) covering a long gradient of intensification. The two principles concerning economy of inputs and reduction of pollution emerged in nearly all the case studies, a finding that can be explained by the economic and regulatory constraints affecting animal production. Integrated management of animal health was seldom mobilized, as alternatives to chemical drugs have only recently been investigated, and the results are not yet transferable to farming practices. A number of ecological functions and ecosystem services (recycling of nutrients, forage yield, pollination, resistance to weed invasion, etc.) are closely linked to biodiversity, and their persistence depends largely on maintaining biological diversity in agroecosystems. We conclude that the development of such ecology-based alternatives for animal production implies changes in the positions adopted by technicians and extension services, researchers and policymakers. Animal production systems should not only be considered holistically, but also in the diversity of their local and regional conditions. The ability of farmers to make their own decisions on the basis of the close monitoring of system performance is most important to ensure system sustainability.
Abstract The agricultural world today is dominated by a few domesticated mammal species, that is, animals modified from their wild ancestors through selective breeding in captivity for traits beneficial to human usages. As a result, a clear dichotomy exists between wild (from hunting) and domesticated mammals (produced in farms) used for human consumption. Similar to agriculture, aquaculture is often viewed as the only solution that can provide more fish products given that harvesting wild stocks have reached an upper limit. Aquaculture is considerably younger than agriculture relying on natural sources to farm numerous species. To better describe the diverse strategies for fish production, we propose a new classification comprising five levels of ‘domestication’ with 1 being the least to 5 being the most domesticated. Our classification places 70% of the 250 farmed finfish species recorded in the 2009 FAO database into levels 1, 2 and 3 representing a transitory form of fish production dependent on the availability of the wild resource. In contrast, only a few species, or more accurately populations, can be considered truly domesticated, similar to cattle or sheep. Based on this classification, two scenarios for the future of aquaculture are discussed: either the industry focuses on few truly domesticated species, similar to the path taken by agriculture, but avoiding its negative impacts or aquaculture proceeds with inter‐specific diversification by focusing primarily on the domestication of native species.
The present study aimed to evaluate the digestion rate and nutritional quality of pig muscle proteins in relation to different meat processes (aging, mincing, and cooking). Under our experimental conditions, aging and mincing had little impact on protein digestion. Heat treatments had different temperature-dependent effects on the meat protein digestion rate and degradation potential. At 70 °C, the proteins underwent denaturation that enhanced the speed of pepsin digestion by increasing enzyme accessibility to protein cleavage sites. Above 100 °C, oxidation-related protein aggregation slowed pepsin digestion but improved meat protein overall digestibility. The digestion parameters defined here open new insights on the dynamics governing the in vitro digestion of meat protein. However, the effect of cooking temperature on protein digestion observed in vitro needs to be confirmed in vivo.
Bumble bees represent one of the most important groups of pollinators. In addition to their ecological and economic relevance, they are also a highly charismatic group which can help to increase the interest of people in realizing, enjoying and conserving natural systems. However, like most animals, bum- ble bees are sensitive to climate. In this atlas, maps depicting potential risks of climate change for bumble bees are shown together with informative summary statistics, ecological back- ground information and a picture of each European species. Thanks to the EU FP7 project STEP, the authors gathered over one million bumblebee records from all over Europe. Based on these data, they modelled the current climatic niche for almost all European species (56 species) and projected future climatically suitable conditions using three climate change scenarios for the years 2050 and 2100. While under a moderate change scenario only 3 species are projected to be at the verge of extinction by 2100, 14 species are at high risk under an intermediate change scenario. Under a most severe change scenario as many as 25 species are projected to lose almost all of their climatically suitable area, while a total of 53 species (77% of the 69 European species) would lose the main part of their suitable area. Climatic risks for bumblebees can be extremely high, depending on the future development of human society, and the corresponding effects on the climate. Strong mitigation strategies are needed to preserve this important species group and to ensure the sustainable provision of pollination services, to which they considerably contribute.
The qualitative properties of the meat are of major importance for poultry breeding, since meat is now widely consumed as cuts or as processed products. The aim of this study was to evaluate the genetic parameters of several breast meat quality traits and their genetic relationships with muscle characteristics in a heavy commercial line of broilers. Significant levels of heritability (averaging 0.3) were obtained for breast meat quality traits such as pH at 15 min post-slaughter, ultimate pH (pHu), color assessed by lightness L*, redness a* and yellowness b*, drip loss, thawing-cooking loss and shear-force. The rate of decrease in pH early post-mortem and the final pH of the meat were shown to be key factors of chicken meat quality. In particular, a decrease in the final pH led to paler, more exudative and tougher breast meat. The level of glycogen stored in breast muscle estimated by the Glycolytic Potential (GP) at slaughter time was shown to be highly heritable (h2 0.43). There was a very strong negative genetic correlation (rg) with ultimate meat pH (rg -0.97), suggesting a common genetic control for GP and pHu. While breast muscle weight was genetically positively correlated with fiber size (rg 0.76), it was negatively correlated with the level of glycogen stored in the muscle (rg -0.58), and as a consequence it was positively correlated with the final pH of the meat (rg 0.84). This genetic study confirmed that selection should be useful to improve meat characteristics of meat-type chickens without impairing profitability because no genetic conflict was detected between meat quality and meat quantity. Moreover, the results suggested relevant selection criteria such as ultimate pH, which is strongly related to color, water-holding capacity and texture of the meat in this heavy chicken line.
The structural and metabolic characteristics of the pectoralis major (P. major) muscle (i.e., breast muscle) and the quality of the resulting meat were studied in relation to breast muscle fiber development in broiler chickens. Six hundred birds originating from a commercial, grand parental, male heavy line (Hubbard-Europe, Châteaubourg, France) were kept under conventional breeding methods until their usual marketing age of 6 wk. For all birds, the plasma creatine kinase activity and the P. major muscle fiber cross-sectional area (CSA), glycolytic potential, lactate content, pH at 15 min postmortem, as well as the ultimate pH, CIELAB color parameters [lightness (L*), redness (a*), and yellowness (b*)], and drip loss of breast meat, were measured. Increased breast weight and yield were associated with increased fiber CSA, reduced muscle glycolytic potential, and reduced lactate content at 15 min postmortem. Therefore, P. major muscle exhibiting larger fiber CSA exhibited greater pH at 15 min postmortem and ultimate pH, produced breast meat with lower L* and reduced drip loss, and was potentially better adapted to further processing than muscle exhibiting small fiber CSA.
Within a population of Charolais young bulls, two extreme groups of longissimus thoracis muscle samples, classified according to Warner-Bratzler shear force (WBSF) of 55 degrees C grilled meat, were analyzed by 2D-electrophoresis. Muscle analyses were performed on 4 bulls of the "tender" group (WBSF=27.7+/-4.8 N) and 4 bulls of the "tough" group (WBSF=41.2+/-6.1 N), at 3 post-mortem times: D0, samples taken within 10 min post-mortem; D5 and D21, samples kept at 4 degrees C under vacuum during 5 and 21 days. Proteins of muscle samples were separated in two fractions based on protein solubility in Tris buffer: "soluble" and "insoluble". Proteins of both fractions were separated by 2D-electrophoresis. Evolution of spots during the 3 post-mortem times was analyzed by hierarchical classification (HCA). Three clusters of proteins presenting similar evolution profiles provided accurate classification of post-mortem times and showed the translocation of some chaperone proteins and glycolytic enzymes from the soluble fraction to the insoluble fraction between D0 and D5. Cellular structure dismantlement and proteolysis was observed at D21. Effect of group ("tender" vs "tough") on spot intensities was tested by ANOVA. At D0, higher quantity of proteins of the inner and outer membrane of mitochondria was found in the tender group suggesting a more extensive degradation of mitochondria that may be related to the apoptotic process.
Bacterial colonization of biotic or abiotic surfaces results from two quite distinct physiological processes, namely bacterial adhesion and biofilm formation. Broadly speaking, a biofilm is defined as the sessile development of microbial cells. Biofilm formation arises following bacterial adhesion but not all single bacterial cells adhering reversibly or irreversibly engage inexorably into a sessile mode of growth. Among molecular determinants promoting bacterial colonization, surface proteins are the most functionally diverse active components. To be present on the bacterial cell surface, though, a protein must be secreted in the first place. Considering the close association of secreted proteins with their cognate secretion systems, the secretome (which refers both to the secretion systems and their protein substrates) is a key concept to apprehend the protein secretion and related physiological functions. The protein secretion systems are here considered in light of the differences in the cell-envelope architecture between diderm-LPS (archetypal Gram-negative), monoderm (archetypal Gram-positive) and diderm-mycolate (archetypal acid-fast) bacteria. Besides, their cognate secreted proteins engaged in the bacterial colonization process are regarded from single protein to supramolecular protein structure as well as the non-classical protein secretion. This state-of-the-art on the complement of the secretome (the secretion systems and their cognate effectors) involved in the surface colonization process in diderm-LPS and monoderm bacteria paves the way for future research directions in the field.
The formation of a nanometer-size oxide pattern on silicon using a scanning probe microscope (SPM) has been widely reported in the literature. No analytical model has been proposed, however, to explain the variation of the oxide height with both polarization and speed of the SPM tip. In this letter, we explain quantitatively the variation of the oxide height with the polarization and the speed of the tip with a model based on field induced oxidation. Data analysis also allows us to estimate the thermal activation energy of the oxidation process, (∼0.15 eV). This low value is compared with activation energies measured for thermal and plasma oxidation of silicon.
Freshwater fish have been widely introduced worldwide, and freshwater ecosystems are among those most affected by biological invasions. Consequently, freshwater fish invasions are one of the most documented invasions among animal taxa, with much information available about invasive species, their characteristics, invaded regions, invasion pathways, impacts, and management. While existing reviews address specific aspects of freshwater fish invasions, there is still a gaping lack of comprehensive assessments of freshwater fish invasions that simultaneously address pivotal and connected elements of the invasion process. Here, we provide a holistic review, together with quantitative assessments, divided into four major parts: ( a) introduction pathways, ( b) characteristics of nonnative species andinvaded ecosystems that explain successful invasion processes, ( c) invasion impacts and their mechanisms, and ( d) management. We highlight data gaps and biases in the current databases and highlight a basic lack of understanding of several aspects of freshwater fish invasions. In addition, we provide recommendations for future studies.
Abstract Temperate fish species are annual spawners and mainly rely on annually cycling cues (temperature and photoperiod) to synchronise the three main phases of their reproductive cycle, that is, induction (initiation of oogenesis), vitellogenesis and the final stages (including maturation, ovulation and oviposition). This review synthesises how these three phases are controlled by specific temperature and photoperiod variations. The direction of the changes (i.e. decrease or increase) is the most important factor, although the amplitude, rates and timing of variations should also be considered to improve/optimise the quality of reproduction in aquaculture. In addition, we tentatively classified temperate fish species sharing similar temperature and/or photoperiod variation requirements for reproduction into three general functional groups. The first group (salmonids) is induced by increasing photoperiod. Vitellogenesis and the final stages are synchronised by decreasing photoperiod. The second group (percids, moronids and gadids) is induced by decreasing both temperature and photoperiod. A chilling period allows vitellogenesis. Increasing temperatures synchronise the final stages. The third group (cyprinids) is induced by decreasing either photoperiod or temperature. Vitellogenesis is faster at warm temperatures. The final stages require an increase in either photoperiod or temperature. This classification may help future research on the control of reproduction in newly cultured fish species.
Livestock generates high quantities of residues, which has become a major socioeconomic issue for the meat industry. This review focuses on the identification of bioactive peptides (BPs) in animal byproducts and meat wastes. Firstly, the main bioactivities that peptides can have will be described and the methods for their evaluation will be discussed. Secondly, the various origins of these BPs will be studied. Then, the techniques and tools for the generation of BPs will be detailed in order to discuss, in the final part, how peptides could be used and assimilated. BPs possess diverse biological activities and can be strategic candidates for substituting synthetic molecules. In silico potentiality studies are a helpful tool to understand and predict BPs released from proteins and their potential activities. However, in vitro validation is often required. Although BP use is compelled by strict regulations in relation to the field of application, they are also limited by their low bioavailability and bioaccessibility. Therefore, it is important to test peptide stability during gastrointestinal digestion. Protective strategies have been discussed since their use could improve the stability and effectiveness of BPs.
The extracellular matrix (ECM) is present within all animal tissues and organs. Actually, it surrounds the eukaryotic cells composing the four basic tissue types, i.e. epithelial, muscle, nerve and connective. ECM does not solely refer to connective tissue but composes all tissues where its composition, structure and organization vary from one tissue to another. Constituted of the four main fibrous proteins, i.e. collagen, fibronectin, laminin and elastin, ECM components form a highly structured and functional network via specific interactions. From the basement membrane to interstitial matrix, further heterogeneity exists in the organization of the ECM in various tissues and organs also depending on their physiological state. Back to a molecular level, bacterial proteins represent the most significant part of the microbial surface components recognizing adhesive matrix molecules (MSCRAMM). These cell surface proteins are secreted and localized differently in monoderm and diderm-LPS bacteria. While one collagen-binding domain (CBD) and different fibronectin-binding domains (FBD1 to 8) have been registered in databases, much remains to be learned on specific binding to other ECM proteins via single or supramolecular protein structures. Besides theinteraction of bacterial proteins with individual ECM components, this review aims at stressing the importance of fully considering the ECM at supramolecular, cellular, tissue and organ levels. This conceptual view should not be overlooked to rigorously comprehend the physiology of bacterial interaction from commensal to pathogenic species.
The blood-brain barrier (BBB) is a term used to describe the unique properties of central nervous system (CNS) blood vessels. One important BBB property is the formation of a paracellular barrier made by tight junctions (TJs) between CNS endothelial cells (ECs). Here, we show that Lipolysis-stimulated lipoprotein receptor (LSR), a component of paracellular junctions at points in which three cell membranes meet, is greatly enriched in CNS ECs compared with ECs in other nonneural tissues. We demonstrate that LSR is specifically expressed at tricellular junctions and that its expression correlates with the onset of BBB formation during embryogenesis. We further demonstrate that the BBB does not seal during embryogenesis in Lsr knockout mice with a leakage to small molecules. Finally, in mouse models in which BBB was disrupted, including an experimental autoimmune encephalomyelitis (EAE) model of multiple sclerosis and a middle cerebral artery occlusion (MCAO) model of stroke, LSR was down-regulated, linking loss of LSR and pathological BBB leakage.
The water intake of 41 lactating dairy cows managed according to current dairy farm practices was individually and continuously monitored to 1) investigate drinking behavior and 2) determine factors affecting water intake. The cows were housed in a free-stall barn and fed once daily with a corn silage and concentrate-based total mixed ration (48% dry matter content; 20.6 +/- 3.3 kg/d of dry matter intake). Cows were milked twice daily, with a yield of 26.5 +/- 5.9 kg/d. The daily free water intake (FWI) was 83.6 +/- 17.1 L, achieved during 7.3 +/- 2.8 drinking bouts. The drinking bout water intake was 12.9 +/- 5.0 L. Almost three-fourths of the FWI occurred during working hours (0600 to 1900 h). Consumption peaks corresponded to feeding and milking times. More than one quarter of the daily FWI was met during the 2 h after each milking. About 75% of the present cows visited the watering point at least once during the 2 h after the evening milking. It is probable that drinking behavior evolved with lactation, but further studies are required to identify the relationship between lactation stage and drinking behavior. The most relevant factors affecting the daily FWI of lactating cows were best combined according to the following predictive equation: (R(2) = 0.45; n = 41 cows, n = 1,837): FWI, L/d = 1.53 x dry matter intake (kg/d) + 1.33 x milk yield (kg/d) + 0.89 x dry matter content (%) + 0.57 x minimum temperature ( degrees C) - 0.30 x rainfall (mm/d) - 25.65. The results obtained using these equations were in agreement with the equations developed by other researchers.
