The DNA colony hybridization test with the polynucleotide probe for Vibrio parahaemolyticus toxR gene was performed. All 373 strains of V. parahaemolyticus gave positive results, and the strains belonging to four other Vibrio species including Vibrio alginolyticus gave weakly positive results, suggesting that toxR sequence variation may reflect the phylogenetic relationships of Vibrio species. We then established a toxR-targeted PCR protocol for the specific detection of V. parahaemolyticus.
ABSTRACT This article argues that divergent images of community result not from inadequate knowledge or confusion of purpose, but from the location of discourse and action in the context of specific struggles and dilemmas. It supports the view that ‘struggles over resources’ are also ‘struggles over meaning’. It demonstrates the ways in which contests over the distribution of property are articulated in terms of competing representations of community at a range of levels and sites. It suggests that, through the exercise of ‘practical political economy’, particular representations of community can be used strategically to strengthen the property claims of potentially disadvantaged groups. In the policy arena, advocates for ‘community based resource management’ have represented communities as sites of consensus and sustain‐ability. Though idealized, such representations have provided a vocabulary with which to defend the rights of communities vis‐à‐vis states. Poor farmers, development planners, consultants and academics can also use representations of community strategically to achieve positive effects, or at least to mitigate negative ones. Most, but not all, of the illustrations in this article are drawn from Indonesia, with special reference to Central Sulawesi.
Vibrio parahaemolyticus O3:K6 strains responsible for the increase in the number of cases of diarrhea in Calcutta, India, beginning in February 1996 and those isolated from Southeast Asian travelers beginning in 1995 were shown to belong to a unique clone characterized by possession of the tdh gene but not the trh gene and by unique arbitrarily primed PCR (AP-PCR) profiles (J. Okuda, M. Ishibashi, E. Hayakawa, T. Nishino, Y. Takeda, A. K. Mukhopadhyay, S. Garg, S. K. Bhattacharya, G. B. Nair, and M. Nishibuchi, J. Clin. Microbiol. 35:3150-3155, 1997). Evidence supporting a hypothesis that this clone emerged only recently and is spreading to many countries was obtained in this study. Of 227 strains isolated in a hospital in Bangladesh between 1977 and 1998, only 22 strains isolated between 1996 and 1998 belonged to the new O3:K6 clone (defined by the serovar, the tdh and trh typing, and AP-PCR profiles). The O3:K6 strains isolated from clinical sources in Taiwan, Laos, Japan, Thailand, Korea, and the United States between 1997 and 1998 were also shown to belong to the new O3:K6 clone. The clonality of the new O3:K6 strains was also confirmed by analysis of the toxRS sequence, which has been shown to be useful for phylogenetic analysis of the members of the genus Vibrio. The toxRS sequences of the representative strains of the new O3:K6 clone differed from those of the O3:K6 strains isolated before 1995 at least at 7 base positions within a 1,346-bp region. A new PCR method targeted to 2 of the base positions unique to the new O3:K6 clone was developed. This PCR method could clearly differentiate all 172 strains belonging to the new O3:K6 clone from other O3:K6 strains isolated earlier. One hundred sixty-six strains belonging to 28 serovars other than O3:K6 were also examined by the new PCR method. The tdh-positive and trh-lacking strains that belonged to the O4:K68 and O1:K untypeable serovars and were isolated in three countries and from international travelers beginning in 1997 gave positive results. The AP-PCR profiles of these strains were nearly identical to those of the new O3:K6 clone, and their toxRS sequences were 100% identical to that of the new O3:K6 clone. The results suggest that these strains may have diverged from the new O3:K6 clone by alteration of the O:K antigens. In conclusion, this study presents strong evidence for the first pandemicity in the history of V. parahaemolyticus and reports a novel toxRS-targeted PCR method that will be useful in epidemiological investigation of the cases associated with the current pandemic spread.
This book presents a technical review of ecological and life history information on a range of Bornean wildlife species, aimed at identifying what makes these species sensitive to timber harvesting practices and associated impacts. It addresses three audiences: 1) those involved in assessing and regulating timber harvesting activities in Southeast Asia, 2) those involved in trying to achieve conservation goals in the region, and 3) those undertaking research to improve multipurpose forest management. This book shows that forest management can be improved in many simple ways to allow timber extraction and wildlife conservation to be more compatible than under current practices. The recommendations can also be valuable to the many governmental and non-governmental organisations promoting sustainable forest management and eco-labelling. Finally, it identifies a number of shortcomings and gaps in knowledge, which the hope can interest the scientific community and promote further research. This review is, an important scientific step toward understanding and improving sustainable forestry practices for long-term biodiversity conservation. Even in the short term, however, significant improvements can be made to improve both conservation and the efficiency of forest management, and there is no need to delay action due to a perceived lack of information. In the longer term it is expected that the recommendations from this review will be implemented, and that further research will continue to help foster an acceptable balance among the choices needed to maintain healthy wildlife populations and biodiversity in a productive forest estate.
A variety of serovars of the food-borne pathogen Vibrio parahaemolyticus normally cause infection. Since 1996, the O3:K6 strains of this pathogen have caused pandemics in many Asian countries, including Taiwan. For a better understanding of these pandemic strains, the recently isolated clinical O3:K6 strains from India, Japan, Korea, and Taiwan were examined in terms of pulsed-field gel electrophoresis (PFGE) typing and other biological characteristics. After PFGE and cluster analysis, all the O3:K6 strains were grouped into two unrelated groups. The recently isolated O3:K6 strains were all in one group, consisting of eight closely related patterns, with I1(81%) and I5(13%) being the most frequent patterns. Pattern I1 was the major one for strains from Japan, Korea, and Taiwan. All recently isolated O3:K6 strains carried the thermostable direct hemolysin (tdh) gene. No significant difference was observed between recently isolated O3:K6 strains and either non-O3:K6 reference strains or old O3:K6 strains isolated before 1996 with respect to antibiotic susceptibility, the level of thermostable direct hemolysin, and the susceptibility to environmental stresses. Results in this study confirmed that the recently isolated O3:K6 strains of V. parahaemolyticus are genetically close to each other, while the other biological traits examined were usually strain dependent, and no unique trait was found in the recently isolated O3:K6 strains.
Although thermostable direct hemolysin (TDH)-producing Vibrio parahaemolyticus has caused many infections in Asian countries, the United States, and other countries, it has been difficult to detect the same pathogen in seafoods and other environmental samples. In this study, we detected and enumerated tdh gene-positive V. parahaemolyticus in Japanese seafoods with a tdh-specific PCR method, a chromogenic agar medium, and a most-probable-number method. The tdh gene was detected in 33 of 329 seafood samples (10.0%). The number of tdh-positive V. parahaemolyticus ranged from <3 to 93/10 g. The incidence of tdh-positive V. parahaemolyticus tended to be high in samples contaminated with relatively high levels of total V. parahaemolyticus. TDH-producing strains of V. parahaemolyticus were isolated from 11 of 33 tdh-positive samples (short-necked clam, hen clam, and rock oyster). TDH-producing strains of V. parahaemolyticus were also isolated from the sediments of rivers near the coast in Japan. Representative strains of the seafood and sediment isolates were examined for the O:K serovar and by the PCR method specific to the pandemic clone and arbitrarily primed PCR and pulsed-field gel electrophoresis techniques. The results indicated that most O3:K6 tdh-positive strains belonged to the pandemic O3:K6 clone and suggested that serovariation took place in the Japanese environment.
This article examines the production of two classes of goods, textiles and ceramics, in the medieval South Indian empire of Vijayanagara. A general model for the organization of specialized craft production is presented in which productive organization is linked to political/administrative regulation of product manufacture, distribution, and use. Three modes of productive organization are defined: administered production, centralized production, and noncentralized production. Historic documentation is used to examine Vijayanagara textile production, and a centralized productive organization is proposed. Vijayanagara ceramic manufacture is assessed through archeological and ethnographic data, and a noncentralized production system is proposed.
Since assuming the presidency, Rodrigo R. Duterte has “stuck to his guns” in carrying out his campaign pledge to launch a violent anti-drug campaign. Duterte's presidency was preceded by six years of political stability and high growth under the relatively liberal and supposedly reformist administration of President Benigno “Noynoy” S. Aquino, III. What did voters find so appealing about Duterte given that drugs and criminality were not a major national concern until he launched his candidacy? Unlike previous populist politicians in the post-Marcos Philippines, Duterte's strongest support did not come from the poorest voters but rather from the elite and the middle class who most feared for their personal security. Although Aquino was widely perceived to be personally honest, his administration had become “systemically disjunctive” and vulnerable to replacement by violent illiberalism because its narrative of “good governance” had been undermined, its strategic allies were weakened, and liberal institutions discredited. Duterte is an illiberal populist who changed the prevailing political order into an illiberal one through a new law-and-order governing script, new key strategic groups (the communist left and the police), and the quick removal of remaining liberal constraints (particularly in Congress and the Supreme Court). Duterte constructed a strongman political model at the local level before “nationalising” it after his election as president.
Sixty-six strains of Vibrio parahaemolyticus belonging to 14 serotypes were isolated from hospitalized patients in Dhaka, Bangladesh, from January 1998 to December 2000. Among these, 48 strains belonging to four serotypes had the pandemic genotype and possessed the tdh gene. A marker (open reading frame ORF8) for a filamentous phage previously thought to correspond to the pandemic genotype was found to have a poor correlation with the pandemic genotype.
The cheetah (Acinonyx jubatus) has been described as a species with low levels of genetic variation. This has been suggested to be the consequence of a demographic bottleneck 10 000-12 000 years ago (ya) and also led to the assumption that only small genetic differences exist between the described subspecies. However, analysing mitochondrial DNA and microsatellites in cheetah samples from most of the historic range of the species we found relatively deep phylogeographic breaks between some of the investigated populations, and most of the methods assessed divergence time estimates predating the postulated bottleneck. Mitochondrial DNA monophyly and overall levels of genetic differentiation support the distinctiveness of Northern-East African cheetahs (Acinonyx jubatus soemmeringii). Moreover, combining archaeozoological and contemporary samples, we show that Asiatic cheetahs (Acinonyx jubatus venaticus) are unambiguously separated from African subspecies. Divergence time estimates from mitochondrial and nuclear data place the split between Asiatic and Southern African cheetahs (Acinonyx jubatus jubatus) at 32 000-67 000 ya using an average mammalian microsatellite mutation rate and at 4700-44 000 ya employing human microsatellite mutation rates. Cheetahs are vulnerable to extinction globally and critically endangered in their Asiatic range, where the last 70-110 individuals survive only in Iran. We demonstrate that these extant Iranian cheetahs are an autochthonous monophyletic population and the last representatives of the Asiatic subspecies A. j. venaticus. We advocate that conservation strategies should consider the uncovered independent evolutionary histories of Asiatic and African cheetahs, as well as among some African subspecies. This would facilitate the dual conservation priorities of maintaining locally adapted ecotypes and genetic diversity.
The genomes of the O3:K6 strains of Vibrio parahaemolyticus which abruptly emerged in Calcutta, India, in February 1996 and which demonstrated an unusual potential to spread and an enhanced propensity to cause infections were examined by different molecular techniques to determine clonality. No restriction fragment length polymorphism (RFLP) in the gene encoding the thermostable direct hemolysin was observed among the O3:K6 isolates of V. parahaemolyticus. Clonal diversity among the O3:K6 strains became evident by examining the RFLPs of the rrn operons and by the use of pulsed-field gel electrophoresis. Five ribotypes were distinguished among the O3:K6 strains examined, with ribotype R4 constituting the major type. Strains of O3:K6 isolated between June and August 1996 showed different pulsotypes compared to the pulsotypes of strains isolated before and after this period, indicating genetic reassortment among these strains, but those isolated between August 1996 and March 1998 showed identical or nearly similar pulsotypes. It is clear that there is a certain degree of genomic reassortment among the O3:K6 clones but that these strains are predominantly one clone.
AIM: To investigate which category in the "Kihon Checklist" developed by the Japanese Ministry of Health, Labor and Welfare can predict functional decline for community-dwelling elderly people at a 2-year follow up. METHOD: We compared comprehensive geriatric assessment (CGA) between "specified elderly individuals" at high risk of requiring long-term care insurance (LTCI) and "uncertified elderly people" (neither certified under LTCI nor "specified"), and also compared CGA between the risk group and non-risk group, in subcategories of the "Kihon Checklist", such as physical strength, nutrition/oral function, overall low score on questions 1-20, houseboundness, cognitive function, and depression risk. The study population consisted of 527 elderly participants aged 75 years and older in a cross-sectional study, and 382 in a longitudinal study. CGA was assessed for basic and higher functional activities of daily living (ADL), depressive symptoms, and quality of life (QOL). The Student's t-test was used in the cross-sectional study and ANOVA with repeated measures was used in the longitudinal analysis. RESULTS: In the cross-sectional study, the risk group had lower functions in all CGA items than the non-risk group in all subcategories of the "Kihon Checklist." In the longitudinal study, Tokyo Metropolitan Institute of Gerontology Index of Competence scores and its three subscales declined in the risk group both in physical and cognitive subcategories compared with the non-risk group, whereas only one or two subscales of Tokyo Metropolitan Institute of Gerontology Index of Competence declined in "specified" and the other two subcategories of the Kihon Checklist CONCLUSION: In both cross-sectional and longitudinal studies, the assessment of physical strength and cognitive function was more useful to detect frail elderly.
This paper first shows that shares of foreign multinational corporations (MNCs) in the manufacturing sectors of five Asian host economies (Hong Kong, Indonesia, Malaysia, Singapore and Taiwan) were generally large in terms of exports, small in terms of employment and moderate in terms of production. Correspondingly, the average product of labour and export propensities were often significantly higher in foreign MNCs than in local firms. In addition, foreign MNCs tended to be relatively large and to have relatively high average capital productivity, capital intensity, skilled‐labour intensity, R&D intensity, profit rates and import propensities, but relatively low shares of labour compensation in value added, and these differences were also statistically significant in many cases. Differences between wholly‐ or heavily‐foreign plants and foreign plants with lower foreign ownership shares were also significant in many cases and generally in the same direction as the differences between MNCs and local plants noted above. Differences among MNCs by foreign source were generally small and insignificant in Hong Kong. In Singapore, European and US firms tended to be larger and characterized by relatively high average labour productivity, capital intensity, profitability and export propensities, but relatively low shares of labour compensation in value added compared to Japanese and other Asian firms.
Vibrio parahaemolyticus is a Gram-negative, halophilic bacterium isolated from marine environments globally. After the consumption of contaminated seafood, V. parahaemolyticus causes acute gastroenteritis. To initiate infection, a wide range of virulence factors are required. A complex group of genes is known to participate in the pathogenicity of V. parahaemolyticus ; however, to understand the full mechanism of infection, extensive research is yet required. V. parahaemolyticus has become the leading cause of seafood-related gastroenteritis in Japan, the United States and several other parts of the world. In addition, outbreaks caused by the pandemic clone of this organism are escalating and spreading universally. To minimize the risk of V. parahaemolyticus infection and warrant the safety of seafood, collaboration between governments and scientists is required. We herein provide an updated review of the pathogenicity determinants and distribution of V. parahaemolyticus to deliver a better understanding of the significance of V. parahaemolyticus and its host–pathogen interactions.
The social learning theory of policy decisionmaking provides clear expectations for the temporal diffusion of policy through the American states (Gray, 1973; Rogers, 1995; Walker, 1969). But the diffusion of morality policy, with its technical simplicity, potential for high salience, and debate over basic moral values, may be driven by nonroutine decisionmaking. We hypothesize that morality policy will diffuse in patterns that depend on the purity of the morality debate surrounding it and the distribution of citizen values regarding it. We identify three distinctive temporal diffusion patterns for state death penalty policies that vary on these characteristics. We conclude that political decisionmaking driving some morality policy diffusion involves less policy learning, and more concern about majority public opinion, than does nonmorality policy.
This paper describes two nautical discoveries buried c.2000 years ago in the Red River alluvial plain, northern Vietnam. One is part of a logboat with a series of empty mortise and locking-peg holes for plank attachment using loose rectangular tenons. The other, from an infant mortuary house, is a series of re-used 4-m-long timbers with exactly the same locked mortise-and-tenon technology. Both finds are interpreted as having belonged to river-boats like those shown on the sides of Heger 1 (Dong Son) bronze drums. Potentially-related technologies from the Mediterranean and China are also discussed. © 2006 The Authors

Thermostable direct haemolysin of Vibrio parahaemolyticus has been shown to be a major virulence factor. The Kanagawa phenomenon (KP), haemolysis induced by this haemolysin on a special blood agar medium, is strongly associated with clinical strains. We have been studying the expressions of various tdh genes encoding this haemolysin to elucidate the significance of the tdh genes possessed by KP-negative strains isolated from patients. We examined the importance of the promoter sequence variation for expression level of the tdh gene in this study. Only the tdh2 gene, one of the two tdh genes (tdh1 and tdh2) present in a KP-positive strain, was previously shown to be responsible for the haemolytic activity of the KP-positive strain. The tdh1- and tdh2-lacZ fusions were used to determine and analyse the promoter sequence by primer extension and site-directed mutagenesis methods. Two bases (positions -24 and -34) within the determined tdh2 promoter sequence were shown to be mostly responsible for the difference in the promoter strength between the tdh2 and tdh1 genes both in Escherichia coli and in V. parahaemolyticus backgrounds. Representative tdh promoters of KP-negative strains are close to the tdh2 promoter; they differ at position -34 but have the same base at position -24 as the tdh2 promoter. We demonstrated that base substitution of the tdh promoters of KP-negative strains only at position -34 is sufficient to increase the expression of these genes to the KP-positive level. Therefore, the tdh genes of KP-negative strains are considered to be potentially important because they can generate a KP-positive subclone by a point mutation in their promoters.
Application of an immunomagnetic enrichment method selective for Vibrio parahaemolyticus serovar K6 allowed isolation of a strain belonging to the pandemic O3:K6 clone of V. parahaemolyticus from fresh shellfish not implicated in a clinical case in southern Thailand. Arbitrarily primed PCR profiles of this strain, clinical O3:K6 strains isolated from sporadic diarrhea cases in the same area, and a standard pandemic O3:K6 strain were indistinguishable.
The government of (post)socialist Laos has conceded more than 1 million hectares of land—5 percent of the national territory—to resource investors, threatening rural community access to customary lands and forests. However, investors have not been able to use all of the land granted to them, and their projects have generated geographically uneven dispossession due to local resistance. Based on twenty months of ethnographic fieldwork, this article compares how dispossession materialized in eight villages targeted by a Vietnamese rubber plantation and a Chinese pulpwood plantation in southern Laos. I contribute to a nascent literature on the political contingencies of dispossession by showing how extraeconomic forces of expropriation are governed relationally. Developing a Gramscian relational environmental governance framework, I demonstrate how such contingencies are shaped by social and political relations among and internal to state, capital, and community actors, leading to either the extension and solidification or contraction and fragmentation of dispossession as a hegemonic mode of development. In the case at hand, I focus on four sets of decisive relations: (1) corporate–state relations that mediate the capacity of investors to mobilize state powers of land expropriation; (2) the state's discursive framing of socioenvironmental relations between communities and their rural environments, which affects how amenable village territories are to acquisition; (3) community–government relations built on kinship, ethnic, or historical links that villagers can use to lodge effective grievances with the state; and (4) coherent and democratic internal village relations that build community solidarity against plantation development.
